Gestation for rats is generally 3 weeks, but rats have been known to give birth anywhere from 19 to 23 days after mating. If she hasn't given birth by day 25 the pregnant doe needs to be seen by a vet.

if you have deliberately bred your rats and left her with a mal overnight, then the morning you removed her/him from the mating cage will be counted as day 1
if its an accidental mating, so you dont actually know the mating date, then when her nipples are really prominent, she will be within a day or two of birthing, she will be also showing signs of nestng about then also
